  • Streptococcus mutans attracts great interest of oral clinical microbiologists because it plays a crucial role in the development of dental caries. Development and evaluation of easy to perform, fast and reliable typing methods is important in all clinical studies. RAPD-PCR with M13 primer as well as rep-PCRs with ERIC, REP, BOX and (GTG)5 primers were performed according to the PCR protocols described previously. All S. mutans strains were typeable by M13 primer and revealed 26 fingerprints ranging from 3 000 to 350 bp. Rep-PCR reactions with ERIC, REP and BOX primers revealed only a few very week or none DNA products; in contrast to these results rep-PCR with the (GTG)5 primer showed 24 fingerprints ranging from 3500 to 200 bp. The (GTG)5-PCR method revealed more complex fingerprints than RAPD-PCR. The clustering revealed by both methods was different and showed comparable discriminative power. The study was supported by projects GACR 310/09/0657 and 1M0528.
