  • Aim of this study was to evaluate cellular fatty acid analysis for characterization of spirochetes. Strains were isolated from arthropods collected in South Moravia, Czech Republic. Fatty acid methyl esters (FAME) profile was determined for five Borrelia burgdorferi sensu lato (s.l.) strains isolated from Ixodes ricinus ticks, one "Spironema culicis" strain recovered from mosquito Culex pipiens and seven spirochetal strains (non identified) isolated from mosquitoes and blackflies. Analysis was performed using a gas chromatography columm in conjuction with Microbial Identification System Sherlock (MIDI Inc.,Newark, DE,USA). Results obtained on the basis of cluster analysis of FAME profiles showed, that the B.burgdorferi sensu lato isolates could be well separated from other spirochetal isolates. We recommended method used in this study as a useful tool preliminary identification of spirochetes isolated from ticks and dipterans.

  • Úkolem předložené práce bylo posoudit možnost využití analýzy methylesterů mastných kyselin pro charakterizaci spirochét. Spirochéty byly izolovány z členovců sebraných na Jižní Moravě. Profil methyl esterů mastných kyselin (FAME)byl stanoven u 5 kmenů Borrelia burgdorferi sensu lato izolovaných z Ixodes ricinus,jeden kmen Spironema culicisizolovaného z Culex pipiens a 7 kmenů, dosud neurčených, izolovaných z komárů.Analýza byla prováděna plynovou chromatografií spojenou s Microbial Identification System Sherlock (MIDI Inc., Newark, DE, USA). Výsledky získané clustrovou analýzou FAME profilů jasně prokázaly, že Borrelia burgdorferi je možné snadno odlišitelná od ostatních spirochét. Doporučujeme využívat v této práci použitou metodu pro předběžnou identifikaci spirochét izolovaných z klíšťat a dipter. (cs)
