  • Conserved genomic sequences distinctive of Staphylococcus aureus phage species 3A, 11, 77, 187 and Twort, representative of phage serogroups A, B, F, L and D, were identified and characterized. PCR primers designed for the above sequences were used for development of a multiplex PCR assay which enabled us not only to classify all phages of the International Typing Set plus 16 additional phages to species, but also to detect prophages in S. aureus genomes. One to four different prophages were unambiguously detected in experimentally lysogenized S. aureus strains, and substantial variation in prophage content was found in 176 S. aureus clinical strains of different provenance. In addition, by using a comparative genomics approach, all the prophages in the S. aureus genomes sequenced to date could be revealed and classified to phage species

  • Z konzervativních sekvencí genomů Staphylococcus aureus bakteriofágových druhů 3A, P11-M15, 77, 187 a Twort reprezentujících fágové seroskupiny A, B, F, L a D bylo navrženo 5 párů primerů, jeden pro každý fágový druh. Pro fágový druh 77 reprezentovaný fágy phi13 a phi77 byly navrženy další dva páry primerů umožňující diferenciaci dvou odlišných fágových kmenů serologické skupiny F integrovaných současně v genomu. Byla zavedena metoda multiplex-PCR a uzpůsobena tak, aby umožnila identifikaci fágů těchto druhů a detekci profágů v genomech S. aureus v reakci v jedné zkumavce. U experimentálně lyzogenizovaných kmenů S. aureus 8325-4 fágy phi11, phi47, phi53, phi84, phi85, phi77 a S. aureus 8325 fágem phi77 se touto metodou podařilo detekovat současně jednoho až čtyři odlišné profágy. Metoda umožňuje přiřadit k fágovému druhu a současně k serologické skupině 23 fágů Mezinárodní standardní řady pro fagotypizaci. Také všechny profágy odhalené v sekvencovaných genomech S. aurues (8325, Mu50, N315, MW2, COL, M (cs)
