  • Celobuněčné lyzáty a membránové frakce bakterie Paracoccus denitrificans byly analyzovány pomocí dvourozměrné elektroforézy s imobilizovanými pH gradienty s cílem charakterizovat rozdíly v expresi proteinů během růstu za aerobních a několika typech anaerobních podmínek (s dusičnanem, dusitanem a oxidem dusným). Komparativní obrazová analýza 2-D gelů odhalila několik skupin z celkového počtu 800 proteinových skvrn, které byly charakteristicky indukovány anebo potlačeny v odpovědi na daný terminální akceptor elektronů. Také respirační inhibitor azid vykazoval významné ovlivnění proteinových profilů. Jelikož však většina proteinů ovlivněných azidem v buňkách rostlých aerobně není totožná s proteiny v buňkách rostlých anaerobně na dusitanu, domníváme se, že azid účinkuje převážně nepřímo prostřednictvím dalších buněčných signálů. Omezená informace o genomu studované bakterie neumožnila identifikovat více než 8 fyziologicky významných proteinů. Veřejně dostupná 2-DE databáze P.denitrificans je dostupná na (cs)
  • Two-dimensional gel electrophoresis with immobilised pH gradients was carried out on total cell lysates and membrane fractions of Paracoccus denitrificans with the aim to characterise differences in protein expression during growth under aerobic and various anaerobic conditions (with nitrate, nitrite or nitrous oxide). Comparative image analysis of the protein pattern revealed several subgroups of the total 800 protein spots resolved that were characteristically induced or repressed in response to individual electron acceptors. The respiratory inhibitor azide also exerted a profound influence upon cellular protein composition. However, since most of the proteins showing an altered expression pattern in cells growing on oxygen differed from those in cells growing on nitrite, we suppose that azide acts mainly indirectly, possibly by influencing other cellular signals. Limited information on the P. denitrificans genome has precluded the identification of more than 8 protein spots as yet. A public accessi
