Description
| - Two strains of human fungal pathogen Cryptococus neoformans and three strains of oportunistic pathogen Aureobasidium pullulans were examined by rhodamine-phalloidin and DAPI staining after paraformaldehyde fixation and Triton X-100 treatment to identify actin cytoskeleton structures and nuclei.Uninuclear C. neoformans cells revealed actin cables oriented to developing bud, actin patches accumulated at growing bud and actin ring in the neck region between mother and daughter cells. In contrast, opportunistic pathogen A.pullulans did not reveal actin cables or patches, while actin rings were seen in multinuclear cells.However, all cells revealed diffuse F-actin cytoplasmic staining, accumulated at tips of young hyphae that may represent simple actin cytoskeleton enabling variable morphogenetic changes. Occurence of multinuclear cells in absence of actin cables supports our hypothesis on the role of actin cables in positioning of cell nucleus.
