  • Haloperidol je neuroleptikum účinné v léčbě různých psychiatrických onemocnění. Tato léčba často vyvolává srdeční arytmie. Metodou whole cell patch-clamp jsme zkoumali vliv haloperidolu na srdeční sodíkový, draslíkové a vápníkový proudy. Byla použita 10 microM koncentrace haloperidolu. U kardiomyocytů izolovaných z dospělých srdečních komor potkana haloperidol inhiboval 95% sodíkového proudu (INa) aktivovaného 40 ms napěťovými pulzy z -75 mV do -20 mV. Blokáda přechodného proudu z buňky (Ito) a proudu na konci pulzu (IK,end) byly měřeny 300 ms napěťovými pulzy z -75 mV do +40 mV. Haloperidol způsobil 80% blokádu Ito a 37% blokádu IK,end, vždy reverzibilní. Dále jsme zkoumali vliv haloperidolu na CaV1.2 L-typ kalciového kanálu přechodně exprimovaný v HEK 293 buňkách. Haloperidol inhiboval zhruba 58% amplitudy měřené na vrcholu IV křivky. Inhibice byla reverzibilní a napěťově závislá. Zvýšení amplitudy depolarizačních pulzů silně zvýšilo velikost ihnibice proudu.   • Haloperidol is a neuroleptic agent effective in treatment of various psychiatric diseases. Cardiac arrhythmias have been associated with this therapy. We have investigated effects of haloperidol on cardiac sodium, potassium and calcium channels using whole cell patch-clamp method. 10 microM concentration of haloperidol was tested. In cardiomyocytes isolated from adult rat ventricles, haloperidol inhibited 95% of sodium current (INa) activated by 40 ms voltage pulses from -75 mV to -20 mV. The block of the transient outward potassium current (Ito) and current at the end of the pulse (IK,end) were measured by 300 ms voltage pulses from -75 mV to +40 mV. Haloperidol caused 80% block of Ito and 37% block of IK,end. In all cases the block was reversible. Further, we have investigated CaV1.2 L-type calcium channel transiently expressed in HEK 293 cells.
