Description
  • The MALDI-TOF MS is an ideal method for routine identification of proteins separated by two-dimensional polyacrylamide gel electrophoresis (2D-P AGE). However, in some cases the outputs from peptide mass fingerprinting (PMF) do not enable unambiguous identification of proteins. In such situation, determination of a partial amino acid sequence of the target protein would be extremely helpful in the PMF identification. Sequencing using MALDI-TOF post-source decay (PSD) usually generates complex spectra and the fragmentation is not complete. PSD sequencing with chemically assisted fragmentation allows to avoid these problems. Application of both methods in identification of proteins of Francisella tularensis is presented.

  • MALDI-TOF MS je ideální metodou pro rutinní identifikaci bílkovin separovaných dvojrozměrnou elektroforézou na polyakrylamidovém gelu (2D-PAGE). Nicméně v některých případech výsledky z fingerprintingu peptidové hmoty (PMF) neumožní jednoznačnou identifikaci bílkovin. V takových situacích by bylo určení částečné aminokyselinové sekvence cílové bílkoviny u PMF identifikací velmi prospěšné. Sekvenace za pomoci MALDI-TOF post-source decay (PSD) obvykle vytváří komplexní spektra a fragmentace není dokončena. PSD sekvenace spolu s chemicky podporovanou fragmentací umožňuje se vyhnout těmto problémům. V následující části tohoto sdělení je prezentována aplikace obou metod při identifikaci bílkovin z Francisely tularensis. (cs)
