  • Cílem této studie bylo ukázat odlišný účinek pre a postsynaptických serotoninových-1A (5-HT1A) receptorů na psychóze podobné chování způsobené podáním antagonisty NMDA receptoru, dizocilpinu (MK-801). Samci potkana kmene Wistar obdrželi dvě dávky (0.025 mg/kg a 1 mg/kg) agonisty 5-HT1A receptoru 8-OH-DPAT (8-hydroxy-2-(di-n-propylamino)tetralin) a MK-801 (0,1 mg/kg a 0,3 mg/kg). Měřili jsme zpracování sensorimotorických informací pomocí testu prepulzní inhibice úlekové reakce (PPI) a lokomoční aktivitu potkanů. Našli jsme odlišný účinek nízké a vysoké dávky 5-HT1A agonisty na deficit v PPI a hyperlokomoci u habituovaných potkanů způsobené MK-801. Nízká dávka 8-OH-DPAT, která aktivuje přednostně presynaptické 5-HT1A receptory, zlepšila deficit v PPI a hyperlokomoci u habituovaných potkanů po podání MK-801 (0.1 mg/kg). Nicméně, vysoká dávka 8-OH-DPAT, která aktivuje oba pre- i postsynaptické 5-HT1A receptory, snížila PPI a zvýšila lokomoční aktivitu po podání nízké dávky MK-801. Podání 8-OH-DPAT samotné (cs)
  • The purpose of the present study was to investigate the opposite effect of the pre- and postsynaptic serotonin-1A (5-HT1A) receptors on the psychotic-like behavior induced by a non-competitive antagonist of the NMDA receptor, dizocilpine (MK-801). Male Wistar rats received two doses (0.025 mg/kg and 1 mg/kg) of 5-HT1A receptor agonist 8-OH-DPAT (8-hydroxy-2-(di-n-propylamino) tetralin) and/or MK-801 in two different doses, 0.1 mg/kg or 0.3 mg/kg.
