  • The aim of this study was to characterize a model of immunosuppression in rabbits, based on changes in the lymphocyte subset distribution and changes in proliferative capacity of lymphocytes 1, 3 and 7 days after the administration of 2 mg/kg dexamethasone phosphate (DXP) three times at 6-h intervals. In blood, neutrophilia and lymphopenia without leukocytosis was detected. One day after DXP administration the absolute numbers of all lymphocyte subsets decreased in the blood, whereas in bone marrow, absolute numbers of all lymphocyte subsets increased significantly. In the thymus, DXP led to a marked reduction of numbers of CD4+CD8+ thymocytes. The proliferative capacity of lymphocytes was lower in the blood and spleen only on day 1, no changes were detected in lymph nodes or in bone marrow. The results demonstrate that this DXP application protocol is useful for the experimental induction of short-lasting immunosuppression in rabbits.

  • Cílem studie bylo charakterizovat model imunosuprese u králíka na základě změn v distribuci subpopulací lymfocytů a změn v jejich proliferační kapacitě 1, 3 a 7 dní po aplikaci 2 mg/kg dexamethazon-fosfátu (DXP) třikrát v 6-hodinových intervalech. V krvi byla detekována neutrofilie a lymfopenie bez leukocytózy. Jeden den po aplikaci DXP, všechny subpopulace lymfocytů klesly v krvi, zatímco v kostní dřeni se absolutní počet všech lymfocytárních subpopulací signifikantně zvýšil, kromě CD79alfa+ buněk, jejichž počet stoupl pouze relativně. Efekt DXP na lymfocyty ze sleziny a mezenteriální a popliteální mízní uzlinu nebyl tak výrazný. V thymu, DXP vyvolal výraznou redukci relativního i absolutního počtu CD4+CD8+ thymocytů. Proliferační kapacita lymfocytů byla nižší pouze první den v krvi a slezině, žádné změny nebyly detekovány v mízních uzlinách a kostní dřeni. Výsledky ukazují, že takovýto protokol aplikace DXP je použitelný pro experimentální indukci krátkodobé imunosuprese u králíka. (cs)
