| - Five commercially available ELISA tests for the detection of antibodies against Mycobacterium avium subsp. paratuberculosis in bovine serum were evaluated using sera from paratuberculosis-free and infected dairy cattle. Sensitivity (Se) and specificity (Sp) were estimated after determination of the cut-off (TG-ROC analysis or the cut-off values recommended by the manufacturers, respectively). When the do cut-off values were applied, the five ELISA tests showed sub-optimal Se and Sp. Adopting the cut-offs recommended by the manufacturers, the Sp of 4 ELISA increased. Se was significantly higher in clinically diseased than in latently infected cattle. Calculation of the positive and negative predictive values indicated that, depending on the test, a considerable proportion of false positive and false negative results have to be expected. Therefore, the suitability of antibody detection for the diagnosis of paratuberculosis in individual animals is questioned.

- Pět komerčně dostupných ELISA kitů pro detekci protilátek proti Mycobacterium avium subsp. paratuberculosis v bovinním séru bylo hodnoceno s využitím sér ze skotu zdravého a trpícího paratuberkulózou. U všech testů byla stanovena citlivost (Se) a specificita (Sp), a to po stanovení prahové hodnoty - %22cut off%22 (pomocí TG-ROC analýzy nebo %22cut off%22 doporučovaná výrobcem). Při aplikaci %22cut off%22 pomocí TG-ROC analýzy vykazovalo všech pět ELISA testů sub-optimální Se a Sp. Naproti tomu při použití %22cut off%22 postupu doporučeného výrobcem vzrostla specificita u 4 ELISA testů. Se byla výrazně vyšší u zvířat v klinické fázi než v latentní fázi onemocnění. Na základě výpočtů předpokládaných pozitivních a negativních výsledků lze očekávat, výrazné množství falešně pozitivních a falešně negativních výsledků. Podle těchto výsledků je tedy vhodnost použití metod založených na detekci protilátek pro identifikaci paratuberkulózy u zvířat diskutabilní. (cs)
