| - Vybrané králíčí polyklonální protilátky proti lidským antigenům byly použity jako příklad nástroje pro studie struktury a vývoje imunitního systému u veterinárních druhů. Směs F(ab')2 fragmentů protilátek proti lidskému Ig kappa-FITC/anti-Ig lambda-RPE byla použita pro dvoubarevnou a, v kombinaci s křížově reagující protilátkou HM-57 proti antigenu CD79alfa, trojbarevnou průtokovou cytometrii B lymfocytů periferní krve psa, kočky, skotu a prasete. Králičí protilátka proti lidskému laktoferinu byla použita pro vizualizaci neutrofilních granulocytů psa, prasete a skotu a byla naržena aplikace dvoubarevné imunofenotypizace psích granulocytů. Dále byla prokázána použitelnost protilátky proti CD3 a TdT pro studie lymfopoézy. Práce jako celek dokazuje, že pečlivě vybrané polyklonální protilátky dostupné na trhu mohou vykazovat širokou křížovou reaktivitu a jsou použitelné pro aplikace ve veterinárním výzkumu. (cs)
- We have used selected rabbit anti-human polyclonal antibodies as an example of useful tools for studies on immune system structure and development in veterinary species. The cocktail of anti-human Ig kappa-FITC/anti-Ig lambda-RPE F(ab')2 fragments was used for two-colour and, in combination with the cross-reactive anti-CD79alpha monoclonal antibody HM-57, for three-colour flow cytometry of canine, feline, bovine and porcine peripheral B-cells. Rabbit anti-human lactoferrin-FITC F(ab')2 fragment was used for visualizing neutrophils in dogs, pigs and cattle and an application for two-colour immunophenotyping of canine granulocyte subsets has been designed. Affinity isolated rabbit anti-human CD3 and anti-human TdT have been shown to represent a ready-to-use tool for in situ studies on primary lymphopoiesis. Altogether, our study show that carefully selected polyclonal antibodies available on the market may possess broad cross-reactivity with applications in veterinary research
