  • Většina vědeckých prací na poli geneticky modifikovaných organismů (GMO) prezentuje detekční metody pro identifikaci GM kukuřice, sóji, řepky nebo bavlny. Ostatním modifikacem, jako jsou např. brambory, je věnována jen menší pozornost. Cílem této práce bylo vyvinout jednoduchou a levnou PCR metodu pro detekci ac2 genu kódujícího antimikrobní peptid z amarantu, který byl použit pro transformaci bramboru. Podstatou metody byla duplex PCR, která umožňovala identifikaci jak ac2 genu, tak specifického genu bramboru v jedné reakci. Součástí metodiky byly také plazmidové pozitivní kontroly, které sloužily pro odhalení přítomnosti PCR inhibitorů, falešně pozitivních výsledků a chybné interperetace výsledků během rutinní analýzy. Citlivost metodiky byla testována na plazmidových řadách a byla stanovena na jednu molekulu na reakci. Systém byl otestován na 14 odrůdách bramboru, které byly, pro gen ac2, negativní. (cs)
  • Most of the scientific papers concerning genetically modified organisms (GMO) are focused on the publication of the detection protocols for GM corn, soyabean, rape or cotton. Minor modifications, such as potatoes, attract little attention. This work was based on developing an easy and cheap PCR method for the detection of the ac2 gene, encoding an antimicrobial peptide from amaranth, which was used for transformation of potatoes. The method was based on duplex PCR which enabled identification of both potato and amaranth specific genes in one tube. The method was enhanced by using positive plasmid controls which were able to reveal presence of inhibitors, reduce the incidence of false negative results, and wrong interpretation during analysis of routine samples. PCR sensitivity using plasmid DNA was determined to be one molecule per reaction. The system was used to find the ac2 gene in 14 variants of potatoes, which were all negative for the inserted ac2 gene from amaranth.
