  • In Salmonella enterica serovar Typhimurium, rrtT retron reverse transcriptase, which is part of St85 retron, is quite ubiquitous and is located in the thdF - yidY intergenic region. In this study we show that rrtT is relatively unstable in multidrugresistant, Salmonella genomic island 1 positive strains. Out of 365 field strains, 55 were free of retron. We propose that intramolecular transposition of IS6100 followed by homologous recombination was responsible for the excision. Excision of retrontogetherwith the right end of SGI1 may also result in its stabilization in the S. typhimurium genome. Experimental deletion of rrtT resulted in an accelerated course of infection in orally infected mice. Since the retron excision occurs exclusively inmultidrugresistant S. typhimurium, it cannot be excluded that such strains may increase their virulence in the future.

  • U kmenů Salmonella enterica serovar Typhimurium nacházíme často retron reverzní transkriptázu (rrtT). Ta je součástí retronu St85, který je lokalizován v oblasti mezi geny thdF - yidY. V této studii jsme zjistili, že u multirezistentních kmenů S.Typhimurium, které nesou v genomu Salmonella genomic island 1, je retron nestabilní. Popsali jsme způsob intramolekulární transpozice IS6100, následované homologní rekombinací, která by mohla vést k deleci retronu. Tato delece retronu spolu s pravýmkoncem SGI1 způsobuje mimo jiné stabilizaci SGI 1 v genomu. Pozorovali jsme zrychlený průběh infekce u myší, infikovaných kmenem se záměrnou delecí rrtT. Protože se tato delece přirozeně vyskytuje jen u multirezistentních kmenů, nedá se vyloučit, že tytokmeny mohou díky této deleci v budoucnosti zvýšit svojí virulenci. (cs)
