| - To determine the suitability of recently derived PCR diagnostic marker (YLM) in marker-assisted breeding of BYDV-resistant spring barley lines, spring barley cultivars differing in response to infection with BYDV in field trials and F3 lines of the Atla s68/Jaspis cross seggregating for Yd2 gene were used for analyses. Using this marker, the presence of Yd2 gene was detected in the resistant cultivars Atlas 68, Brea´S/Ben and the CIMMYT lIne GIza 121/Pue. Yd2 signal was not detected in the moderately resistant cultivars Malvaz and Atribut and the five susceptible cultivars tested. Analyses of F3 plants revealed that this codominent PCR diagnostic merker, able to distinguish homozygous and heterozygous condition, may be a valuable tool fo detecting resistant materials also on individual plant basis. Besides, diagnosis by YLM marker was found less costly and time consuming in comparison to field tests.
