| - Preliminary studies were performed to prove, whether sequence-based primers could be used for gliadin/glutenin leci identification instead of polyacrylamide or starch electrophoresis, The possibility of evaluation of DNA-based markers using direct temp late were studied showing that a segment of tissue is usable for PCR reaction in wheat tissue. Different sets of primers specific for gliadin/LMW glutenin loci were used to verify the reliability of the reaction. Segment of leaf tissue is suitable, unex oensive template, whereas coleoptile did not work for this purposes. Two sets of primers derived from gamma-gliadin pseudogne were used to analyse a set of wheat cultivars with standard gliadin alleles. Using F1xR and F2xR primer combinations, 13 differ ent band were recorded, which made differentiation of 12 groups possible. Such method could be therefore a simple and alternative method for their verification.
