  • Background: Sodium channels (SC) in mites and insects are target sites for pesticides including pyrethroids. Point mutations of SC gene were reported to change structural conformation of the protein and sensitivity to pesticides. To find mutations in SC gene of Varroa destructor (VmNa) we analyzed VmNa gene sequences available in GenBank and prepared specific primers for amplification of two fragments harboring the regions coding for (i) IIS4-IIS6 in positions 2837-3337 and (ii) IIIS4-3´terminus in positions 4737-6500 according to VmNa cDNA sequence AY259834. Results: Sensitive and resistant populations did not differ in amino acid sequences of the IIIS4-3´terminus VmNa region. The differences were found in the fragment IIS4-IIS6: In resistant population, mutation C(3004)>G resulted in substitution L(1002)> (codon ctg > gtg) in the position equivalent to L925 in house fly in the linker connecting segments IIS4 and IIS5. Additionally, mutation F(1052) >L (codon ttc > ctc) in the position equivalent to F975 in house fly in the linker connecting segments IIS5 and IIS6 was detected in both resistant and sensitive populations with prevailing incidence in resistant population. Conclusion: All individuals surviving the tau-fluvalinate application in bioassay harbored L(1002)>V mutation, while L(1002) sequence was found in dead individuals from sensitive population as well as dead individuals from resistant population. Further research is necessary to clarify the role of mutation F(1052)>L in the linker connecting the transmembrane VmNa segments IIS5-IIS6 in resistance of V. destructor to tau-fluvalinate.
