  • Seeds of seven weed species suspected to be resistant to atrazine and one to imazapyr were collected along railroads in different parts of the Czech Republic in 2000-05. Greenhouse and laboratory experiments were performed to confirm herbicide resistance. A region of the psbA gene and two regions of the ALS gene were sequenced and compared between the resistant and susceptible biotypes. The psbA gene of the 7 resistant weed biotypes was mutated at residue 264 with substitution of glycine for serine. The seven susceptible weed biotypes sequences were wild-type at this residue. Resistance to imazapyr in the kochia biotypes was conferred by a leucine for tryptophan substitution at residue 574 of ALS. Identification of the amino acid substitution confirmed that the herbicide resistance in all investigated weed species was site-of-action mediated.

  • Semena sedmi plevelných druhů podezřelých z rezistence vůči atrazinu a jednoho druhu podezřelého z rezistence vůči imazapyru byla sesbírána v okolí železničních uzlů v různých částech České republiky. Rezistence byla prokázána pomocí skleníkových a laboratorních pokusů. Byla sekvencována jedna oblast genu psbA a dvě oblasti genu ALS a získané sekvence rezistentních a citlivých biotypů byly vzajemně porovnány. Gen psbA rezistentních biotypů plevelů obsahoval v pozici 264 glycin místo serinu. Sedm citlivých biotypů mělo v této pozici serin. Rezistence vůči imazapyru u biotypu bytele metlatého byla podmíněna substitucí tryptofanu leucinem v pozici 574 ALS. Nalezení záměn aminokyselin potvrdilo, že rezistence vůči herbicidům u všech zkoumaných biotypů plevelů byla způsobena změnou místa jejich působení v rostlině. (cs)
