Description
  • The ploidy level and chromosome number were verified in nine Czech and two US populations of kochia (Kochia scoparia (L.) Schrad.) from various sites and with different susceptibility to acetolactate synthase (ALS)-inhibiting and atrazine herbicides. Radicles 5-15 mm in length from pre-germinated seeds of kochia were used for the karyological analysis. None out of eleven examined populations comprised a case where all 50 analyzed roots would have the diploid number of chromosomes (2n=18). A low frequency of tetraploidy (4n=36) was found in the population with multiple resistance to atrazine and ALS inhibitors. Besides diploids a high frequency of mixoplids was recorded. A region of the gene encoding D1 protein of photosystem II was amplified and sequenced in three Czech kochia biotypes to determine the molecular basis for the atrazine resistance. It was found out that the resistance in the %22Bubny%22 and %22Vršovice%22 kochia biotypes was conferred by a glycine for serine substitution at residue 264 of the
