  • Performance characteristics of two alternative analytical methods used for the determination of zearalenone and its metabolites were assessed in presented study. Extraction of ground grains with acetonitrile-water(84/16, v/v) mixture followed by clean-up using GPC BioBeads S-X3 soft gel were employed prior to HPLC (RP column C18). The use of ELISA method allowed direct, highly sensitive measurement of zearalenone levels in crude extracts. For ELISA method, extraction with methanol-water (70/30, v/v) without any clean-up was used. Compared to HPLC/FLD method, the precision of ELISA method was inferior. The second part of study was concerned with toxic potential of two isolates of Fusarium culmorum under field conditions. Analyses of wheat varieties tested for the content of zearalenone were carried out. Levels of zearalenone in wheat inoculated by more toxic isolate (B) ranged from 309,1 to 2162,7 microgram/kg.

  • V předložené studii byla hodnocena využitelnost dvou alternativních analytických metod použitých pro stanovení zearalenonu a jeho metabolitů. Rozemletá zrna pšenice byla extrahována směsí acetonitril - voda (84/16, v/v),extrakt byl přečistěn na gelové kolonce GPC BioBeads S-X3 a podroben HPLC (RP column C18). Aplikace metody ELISA umožnila přímé, vysoce citlivé stanovení obsahu zearalenonu v surovém extraktu (extrakce směsí methanol - voda 70/30, v/v). V porovnání obou metod (HPLC/FLD a ELISA) je posledně jmenovaná méně přesná. V další části studie byl porovnáván potenciál toxicity dvou izolátů Fusarium culmorum v polních podmínkách. Obsah zearalenonu se lišil v různých testovaných uměle inokulovaných odrůdách pšenice a u izolátu B se pohyboval od 309,1 do 2162,7 mikrogramů/kg. (cs)
