Description
  • The aim of the presented grant proposal is to employ novel analytical techniques, LC/TOF-MS and DART, for metabolomic profiling of several sets of plants grown by our COST research partner (Institute of system biology and ecology, Academy of science of the Czech Republic, Nove Hrady) under well controlled conditions in an experimental field. Within the first phase of pilot study, the proposed TOF MS methods will be optimized and validated the subsequent step will focus on the messages obtained by metabolomic profiling of plant homogenates. Based on the collected information, the “database“ of fingerprints detected at different extent of abiotic stress resistance, will be established. Biomarkers, obtained by statistical evaluation of recorded metabolomic profiles, will be used to identify the biological parameters that will be employed to assess the abiotic stress (drought, light, cold etc.) resistance capacity of genetically modified mutants/transgenics and different ecotypes.   • V rámci interdisciplinárního projektu bude zavedena nová strategie indikace resistence rostlin k abiotickému stresu. Pro záznam změn metabolomických profilů bude využita hmotnostní spektrometrie s průletovým analyzátorem.

  • Pro metabolomickou analýzu vzorků listů Arabidopsis thalianPro metabolomickou analýzu vzorků listů Arabidopsis thaliana byly vyvinuty metody využívající kapalinovou chromatografii s hmotnostní spektrometrii a novou ionizační techniku pro přímou analýzou v reálném čase. Statistická analýza získaných metabolomických záznamů umožnila rozlišit rostliny dle jejich tolerance vůči nízkým teplotám. (cs)
