The aim of the project is the identification and characterization of transcription factors involved in the control of cell wall synthesis in developing male gametophyte. A key prerequisite is to achieve an affirmation of their transcription factor activities (localisation in the nucleus, binding to DNA, modification of gene expression). The second objective is to describe regulons of relevant factors, to find controlled metabolic pathways and their possible overlaps and in the longer perspective to investigate the possible coordinated action of examined factors. Cílem projektu je identifikovat transkripční faktory zodpovědné za regulaci syntézy buněčné stěny pylového zrna, popsat jejich funkci a ověřit jejich koordinované působení.
Byla provedena detailní expresní analýza promotorů zkoumaných genů účastnících se kontroly syntézy buněčné stěny pylového zrna. Experimentálně jsme potvrdili jejich expresní profil a lokalizaci. Byla ukončena komplexní analýza příslušných mutantních T-DNA linií nesoucích mutace v kaniddátských genech, jež se významným způsobem podílejí na vývoji pylu u huseníčku včetně syntézy buněčné stěny. We performed detailed analysis of the tested promoters of genes involved in the control of cell wall synthesis in pollen grains. We experimentally confirmed their expression profile and localization. We also completed the comprehensive analysis of T-DNA mutant lines carrying mutations in candidate genes that are significantly involved in pollen development including cell wall synthesis. (en)
