Description
| - Obesity and especially excessive accumulation of adipose tissue in the abdominal region is associated with an increased risk of developing metabolic syndrome, a cluster of diseases including glucose intolerance, hypertension, and dyslipidemia, that predisposes to the development of type II diabetes and cardiovascular disease. The characteristic feature associated with obesity and the pivotal factor in the pathogenesis of type II diabetes is insulin resistance. It may be detected many years before the clinical onset of hyperglycemia and most likely results from lipotoxicity due to increased accumulation of lipids in the peripheral tissues in the setting of enhanced release of fatty acids from hypertrophic fat cells. Besides this mechanism, also various adipocyte-secreted hormones (adipokines) are involved in the modulation of systemic insulin sensitivity. Systemic low-grade inflammation has been proposed to have an important role in the pathogenesis of obesity-related insulin resistance. Obesity-assoc (en)
- Cílem projektu je testovat hypotézu, že modulace energetického stavu tukových buněk vede ke změně sekreční aktivity tukové tkáně a tím i ke změnám v inzulínové citlivosti, glukózového a lipidového metabolismu celého organismu.

| - V rámci řešení projektu jsme prokázali (i) významnou roli AMPK, senzoru energetického stavu buňky, v benefičních účincích n-3 mastných kyselin (n-3 PUFA), (ii) aditivní účinek kombinovaného podávaní n-3 PUFA a antidiabetik - thiazolindionů na inzulínovou citlivost organismu. (cs)
- Within the project we have demonstrated (i) the important role of AMPK, a cellular sensor of energy status, in beneficial effects of n-3 fatty acids (n-3 PUFA), (ii) the additive effects of the combination treatment of n-3 PUFA and antidiabetic agents – thiazolinediones on insulin sensitivity.
