Description	The aim of the proposed study os to characterize the relationship between the induction of alloreactive T lymphocytes and their capacity to produce Th1 or Th2 cytokines in mixed lymphocyte cultures (MLC) and in patients after kidney transplantation. The frequencies of IFN gamma and IL-4 producing cells will be evaluated as a prognostic factor in kidney transplant patients with respect to the occurence of rejection episodes, allograft function and survival. Studied will be the direct influence of Th1 and Th2 cytokines on renal epithelial cells and the possibilities for modulation of this effect. (en) Projekt si klade za cíl charakterizovat vztah mezi výskytem alloreaktivních T lymfocytů a jejich kapacitou produkovat Th1 nebo Th2 cytokiny ve směsných lymfocytárních kulturách (MLC) a u pacientů po transplanatci ledvin. Výskyt IL-4 a IFN gama produkujících buněk bude posouzen jako prognostický faktor u pacientů s transplantacemi ledvin s ohledem na vznik rejekčních epizod, funkci a přežití transplantovaného štěpu. Na modelu tkáňových kultur bude sledován přímý efekt Th1 a Th2 cytokinů na renální epitel a možnosti modulace účinku Th1/Th2 cytokinů in vitro.
