  • The mechanisms, which may play a role in the development of tissue resistance to insulin action, will be studied in experimental models of insulin resistance, ie, in the non-obese line of heeditary hypertriglyceridemic rats and in the obese line of KoletThe parameters to be evaluated include substrate competition of glukose and NEFA, activation of PPAR transcription factors, and expression of lipoprotein and hormone-sensitive lipase. The effect of specific intervention by thiazolidinedione derivatives, The studies should furnish new data and information on the pathophysiological mechanisms of insulin resistance and to provide hints on the potential for beneficial interventions.   • U experimentálních modelů insulinové resistence, u neobesní linie hereditárně hypertriglyceridemických potkanů a obesní linie Koletského potkanů, budou sledovány mechanismy, které se mohou uplatnit v rozvoji resistence tkání k účinku insulínu. Sledována bude substrátová kompetice glukosy a NEMK, aktivace transkripčních faktorů PPAR a exprese lipoproteinové a hormon.sensitivní lipasy. Dále bude sledován vliv intervence thiazolidindionovými deriváty, fibráty a dehydroepiandrosteronem. Studie by měly přispět k získání poznatků o patofysiologických mechanismech insulinové resistence a naznačit možnosti pozitivní intervence.
