Description
| - Our preliminary experiments demonstrate that in eukaryotic cells novel posttranslational modification of proteins, methylation of glutamic acid (E), found in the P-TEFb binding domain of Hexim1, directs association of P-TEFb with 7SK RNP and thus kinase activity of cdk9. This project will in detail characterize the functional importance of this modification for the regulation of kinase activity of cdk9 furthermore, we plan to identify the first eukaryotic methyltransferase of glutamic acid and define its role in the regulation of association of P-TEFb with 7SK RNP. In the last stage of the project, we plan to isolate the protein complexes associated with cyclin T1, cyclin T2 and cyclin K. The protein composition of individual cyclin complexes may be the very first step in answering the intriguing question regarding the physiological role of cyclinT1, cyclin T2 and cyclin K. (en)
