The objective of the proposed work is to understand the relationship between structure and function for the E. coli protein WrbA, the founding member of a widespread and highly conserved family of proteins. Although WrbA is related to the well-studied bacterial FMN-dependent flavodoxin electron carriers and eukaryotic FAD-dependent oxidoreductases, it presents significant distinctions from both, and has a completely unknown physiological role. The very wide phylogenetic distribution of WrbA homologs suggests an important metabolic role that may reveal unexpected evolutionary and ecological relationships among distant organisms. New hypotheses that follow from our previous work on WrbA structures will be evaluated in a comparative structure/function framework with its better-understood relatives. Hlavním cílem projektu bude snaha o celkovou charakterizaci nové rodiny WrbA proteinů na molekulární úrovni a výzkum biologické funkce těchto proteinů.
The project was focused on general characterization of a new family of WrbA proteins on molecular level as well as at description of their unknown biological function. Basic on successful solving of particular aims is possible to state that the project was solved with big scientific success. The results were published in 6 publications. (en)
