Description
  • The effect of the working conditions on the production rate and on the recovery yield of preparative separations by High- Performance Liquid Chromatography will be investigated to develop working approach allowing increasing separation productivity in preparative chromatography. Multicomponent distribution data between the mobile and the stationary phase will be measured for various types of compounds in sample mixtures under varying experimental conditions. Various equations suitable to describe the distribution isotherms and various kinetic models of mass transport between the phases will be compared to be used for predicting band profiles in preparative separations on overloaded columns. Effects of the mobile phase, of the sample solvent, of the sample volume and of the profile of the mobile phase gradient on separation will be studied. Optimization strategy will be developed for isocratic and gradient elution on overloaded columns, with special attention to the separation of optical isomers. (en)
