Description	Schistosomiasis, one of the world`s most important parasitic diseases, is caused by Schistosoma bloodflukes. Cathepsins that digest host blood proteins in the parasite gut represent a promising intervention target for the treatment of schistosomiasis. The main goal of the project is the characterization of schistosomal cathepsins and the development of their specific inhibitors as potential chemotherapeutics. The specific aims of the project are as follows: (a) Evaluation of cathepsins as effective targets using RNAi and chemical genomics. (b) Recombinant expression of cathepsins and their biochemical and structural characterization. (c) Rational design of selective inhibitors of cathepsins, and (d) Screening for their potency against the parasite in vitro and in vivo. This interdisciplinary research integrates the areas of structural and functional biochemistry and functional genomics and parasitology. The project includes technology transfer and student training programs. (en) Projekt je zaměřen na výzkum katepsinů z parazitických krevniček rodu Schistosoma. Cílem projektu je charakterizace katepsinů a vývoj jejich specifických inhibitorů jako potenciálních chemoterapeutik pro léčbu schistosomózy. Projekt zahrnuje tyto dílčí cíle: (a) zhodnocení významu katepsinů jako cílových molekul pomocí RNAi a chemické genomiky, (b) přípravu katepsinů pro jejich biochemickou a strukturní charakterizaci, (c) racionální vývoj specifických inhibitorů katepsinů a (d) testování účinnosti inhibitorů in vitro a in vivo. Interdisciplinární projekt umožňuje propojit výzkum v oblasti funkční a strukturní biochemie (pracoviště v ČR) a funkční genomiky a parazitologie (pracoviště v USA). Součástí projektu je výměna technologií a školící program pro studenty.
