Description	Little is known about control of microgametogenesis and even less about the role of formins, conserved actin organizers, during plant development. This project links both research fields, as the Arabidopsis formin mutant fh3 with disruption in the AtFH3 locus, showing an unique pollen-specific phenotype, has been isolated in our laboratory. I intend to characterize specific features of mutant plants and also to elucidate the role of formins during pollen development. Research strategy will involve several approaches, with emphasis on genetical studies involving tetrad analysis and crosses of fh3 to other mutant or stably transformed lines. Comprehensive cytological analysis of pollen phenotype will comprise both conventional methods and introduction of novel methodical approach, i.e. immunolocalization of proteins in pollen protoplasts. For this purpose, anti-AtFH3 antibody will be produced and also possible co-localization with cytoskeleton structures will be studied. (en) Tento projekt propojuje dvě velmi zajímavé, avšak málo prostudované oblasti rostlinné biologie, a to studium mikrogametogeneze a studium úlohy forminů jakožto organizátorů aktinového cytosketetu ve vývoji rostliny. V naší laboratoři jsme izolovali mutanta huseníčku fh3 s poškozením v lokusu pro gen AtFH3, který se vyznačuje jedinečným pylově specificým fenotypem, a který bude v rámci tohoto projektu podrobně studován. Dále se pokusím objasnit úlohu forminů při vývoji pylu. Ke studiu pylového fenotypu použiji především genetický přístup, konkrétně tetrádovou anylýzu a křížení fh3 s dalšími mutantními či stabilně transformovanými liniemi. Cytologickou charakteristiku provedu jak pomocí obvyklých postupů, tak za použití nové metodiky, která spočívá v imunochemickém značení proteinů v pylových protoplastech. K tomuto účelu připravím specifickou protilátku proti AtFH3, která bude použita také v experimentech dvojitého značení spolu s jednotlivými složkami cytoskeletu.
