Description
  • Proteins with WD40 motif are directly involved in signal transduction, transcription, pre-mRNA splicing, cytoskeletal organisation, and vesicular fusion and transport. The function of WD40 repeats themselves in particular proteins, insofar as their function is known, consists in protein-protein interaction and in building up functional protein complexes. The aim of this proposal is: 1) to isolate functional WD40 proteins from Thermomonospora curvata and Streptomyces coelicolor A3(2), 2) to investigate the subcellular location of the proteins in mycelium, 3) to identify and to analyse PkwA and WdpA (=WD40 proteins) ligand proteins from T. curvata and S. coelicolor with the help of in vivo and in vitro techniques, 4) to detect protein regions responsible for specific ligand interaction, 5) to explore the regulation of pkwA and wdpA genes, and 6) to determine the role they can play in signal transduction in the process of cell differentiation.   • Proteiny s opakujícím se WD40 motivem se přímo podílejí na přenosu signálů v buňce, transkripci, sestřihu pre-mRNA, uspořádání cytoskeletu, a na vezikulární fúzi a transportu. Funkce samotných WD40 motivů v jednotlivých proteinech, u nichž jsou známy jejich funkce, spočívá v interakci protein-protein a ve formování funkčních proteinových komplexů. Cílem tohoto grantového projektu je: 1)izolace funkčních WD40 proteinů z aktinomycét Thermomonospora curvata a Streptomyces coelicolor A3(2), 2) lokalizace proteinů uvnitř buněk mycélia, 3) identifikace a analýza proteinových ligand PkwA a WdpA (=WD40 proteiny) proteinu z T. curvata a S. coelicolor s použitím in vivo a in vitro technik, 4) určení oblastí proteinových molekul zodpovědných za specifickou vazbu ligandů, 5) studium regulace pkwA a wdpA genů a 6) určení jejich role již mohou hrát při přenosu signálu v procesu diferenciace buněk.

  • Protein PkwA from T. curvata was found in the membrane fraction, and in tips of fast-growing mycelium from the early stage of growth, what confirms its role in signal transduction during this period. Two WD proteins were characterised from Streptomyces. (en)
