Description
  • Study of physiology of insect neurohormones and their effects on the selected taxons of insect pest corresponds with basic principles of the Integrated Pest Management. The aphids (Sternorrhyncha, Aphidoidea) are suitable insect target group for this research because of their evident economic importance and with respect to their hormonally controlled reproductive polyphenism. The aim of this project is to approximate the effect of adipokinetic hormone (AKH) on vitellogenin (Vg) distribution and reproduction of the model species - the pea aphid, Acyrthosiphon pisum Harris. The interactions of the AKH with juvenile hormone (JH) will be studied subsequently. Concentration and distribution of the AKH and Vg in specific morphs of A. pisum will be monitored with the aid of electrophoretic, chromatographic (HPLC) and immunodetection techniques (ELISA and immunohistochemistry). These characteristics will be evaluated under normal physiological conditions and after external application of AKH or JH (JH analogue alternatively). Obtained data will be used for description of relationships between AKH and JH and their effect on vitellogenesis of A. pisum. The last point of the work will focus on the effect of AKH on reproduction of this aphid species that will be expressed by changes in demographic characteristics. (en)
