Description	Corneal transplantation is the most common and successful transplantation in the human medicine. However, a significant part of transplants undergoes an immunological rejection reaction and so far no therapy is able to avert them from this irreversible stage. Therefore, this project is focused on extension of the present knowledge of the mechanisms of allogeneic corneal transplant rejection. For this investigation will be used a model of orthotopic corneal transplantation in mice. The special emphasis will be put on the comparison of effects of immunosuppressive drugs on the prevention of rejection and on the elucidation of their effect on interactions between specific and non-specific immunity cells. These findings will enable the targeted regulationof the effector mechanisms of allotransplantation reaction. A correlation between the expression of surface markers or the expression of iNOS and the clinical stage of transplant will be tested in control and treated groups in various time intervals (en) Transplantace rohovky patří k nejčastěji prováděným a nejúspěšnějším transplantacím v humánní medicine. U části transplantátů však dochází k imunologické rejekční reakci a dosud neexistuje účinná terapie, která by tento ireversibilní stav odvrátila. Proto bude výzkum zaměřen na rozšíření dosavadních znalostí mechanismu rejekce alogenního transplantátu rohovky. Pro toto studium bude využit model ortotopické transplantace rohovky u myši. Zvláštní důraz bude kladen porovnání účinku imunosupresivních látek na prevenci rejekce s cílem zjistit jejich vliv na interakce buněk specifické i nespecifické imunity, a tím umožnit cílenou regulaci efektorových mechanismů alotransplantační reakce. Zjišťována bude korelace mezi expresí povrchových markerů makrofágů, expresí iNOS a klinickým stavem transplantátu v závislosti na časovém intervalu od provedení transplantace u kontrolní skupiny a skupin ovlivněných terapií.
