Description
| - Formins, or FH2 proteins, are an evolutionarily conserved family of actin-organizing proteins exhibiting a great diversity of domain combinations across eukaryotes. Only a small fraction of this variety has been experimentally studied so far. Plants have three groups of formins. For two of them - Class II (where we previously discovered a novel domain related to the PTEN antioncogene), and Class III (recently described by us, found only in lower plants and characterized by the presence of a RhoGAP-like domain) - no experimental report has been published yet. We plan to characterize selected representatives of Class II and Class III formins from Arabidopsis thaliana and Physcomitrella patens, respectively, with respect to their cellular localization, protein-protein interactions, and biological function. The results should contribute to the understanding of the mechanisms governing plant cytoskeletal organization, as well as to grasping the more general problem how combination of conserved domains contributes to the vast diversity of form and function found in eukaryotes. - Forminy (FH2 proteiny) jsou skupina evolučně konzervovaných aktin-organizujících proteinů, které se vyznačují velikou rozmanitostí doménové stavby u různých eukaryot. Jen malá část této diversity byla dosud uchopena experimentálně. Rostliny mají tři skupiny forminů. Pro dvě z nich - třídu II (v níž jsme dříve popsali novou doménu příbuznou antionkogenu PTEN) a třídu III (kterou jsme nedávno objevili u nižších rostlin a pro niž je typická doména příbuzná RhoGAP) - dosud nebyla zveřejněna žádná experimentální data.Naším cílem je charakterizovat vybrané zástupce forminů třídy II a III z modelových rostlin Arabidopsis thaliana a Physcomitrella patens s ohledem na buněčnou lokalizaci, protein-proteinové interakce a biologickou funkci.Výsledky přispějí k pochopení mechanismů řídících uspořádání rostlinného cytoskeletu i k pochopení obecnější otázky, jak může kombinace evolučně starých domén přispívat k rozmanitosti struktur i funkcí eukaryotních buněk.

| - Úspěšné řešení grantu vedlo k získání významných poznatků o souvislostech mezi rostlinnými forminy a dynamikou aktinového cytoskeletu. Výsledky byly publikovány v 3 původních a 3 přehledných sděleních, dále v metodické práci, projekt lze tedy hodnotit jako splněný. Byla obhájena diplomová a disertační práce. Nebyly shledány nedostatky stran dodržování grantových pravidel. (cs)
- The project lead to substantial insights into relationships between plant formins and dynamics of the actin cytoskeleton. The results were published in 3 original papers, 3 reviews and 1 methodological paper, project can thus be evaluated as accomplished. One master and 1 PhD theses were defended. No shortcomings regarding adherence to grant administration rules were found. (en)
