Description
| - The metabolic syndrome represents a cluster of metabolic disorders that increase the risk of type 2 diabetes and cardiovascular disease. The key role is ascribed to dyslipidemia and associated ectopic lipid accumulation in non-adipose tissues such as skeletal muscle, liver, pancreas, and myocardium. This metabolic milieu may induce cellular stress, potentiates oxidative stress and activates inflammatory processes. Using 13C MR spectroscopy, we will study the role of fatty acid transport, their accumulation in tissues, the influence of glucose utilization on glycogen synthesis and other disturbances in the pathogenesis of insulin resistance and its complications in several unique experimental models: a) SHR rats with a deletion or increased expression of fatty acid translocase, b) rats with increased expression of C-reactive protein, c) SHR rats with increased expression of SREBP-1a in the liver, leading to massive steatosis due to the stimulation of fatty acid synthesis de novo, and d) rats with hereditary hypertriglyceridemia. These models will be further analyzed for the possibility of nutritional or pharmacological intervention in their pathology. (en)
- Metabolický syndrom představuje soubor poruch, které zvyšují riziko rozvoje diabetu 2. typu a kardiovaskulárních onemocnění. Klíčovou úlohu má dyslipidémie a s ní spojené ektopické ukládání lipidů do svalů, jater pankreatu a myokardu. Tyto poruchy, představují stres pro buňky a buněčné organely, vedou k inzulinové rezistenci, zvyšují oxidační stres a aktivují zánětlivé procesy. S využitím 13C MR spektroskopie bude v projektu sledována úloha transportu mastných kyselin, jejich akumulace ve tkáních, ovlivnění utilizace glukózy pro syntézu glykogenu a další poruchy uplatňující se v patogenezi inzulinové rezistence a jejích komplikací u unikátních experimentálních modelů: a) potkanů SHR s delecí nebo zvýšenou expresí translokázy mastných kyselin, b) potkanů se zvýšenou expresí C-reaktivního proteinu, c) potkanů SHR se zvýšenou expresí SREBP-1a v játrech, která vede k masivní steatóze v důsledku stimulace syntézy mastných kyselin de novo; d) potkanů s geneticky fixovanou hypertriglyceridémií. U těchto modelů budou dále analyzovány možnosti nutričního a farmakologického ovlivnění.

| - Primární cíle byly dosaženy pouze částečně. Projekt byl zastaven po 3. roce (ze 4 plánovaných) z důvodu neúčasti hlavního řešitele na publikačních výstupech. Panel zaznamenal, že hlavní řešitel není součástí publikací dedikovaných projektu. Prozatím dedikované publikace jen okrajově reflektují cíle či metodiku, jež je v grantu navržena. (cs)
- Primary objectives were achieved only partially. Project was stopped after 3 years (of 4 originally planned) due to the fact that main investigator is not involved in major publication outcome. Panel noticed that main investigator is not a co-author of the dedicated publications and those publications only partially reflect specific aims of the proposal. (en)
