Drug resistance of parasitic helminths in farm animals and wild animals represents the serious world-wide problem in helminthoses therapy. In development of helminth resistance the induction (i.e. increase of activity and expression) of biotransformationenzymes of parasite and host could play important role. From this point of view induction of oxidation and conjugation enzymes have been studied in helminths, but no attention has been paid on reduction biotransformation enzymes although helminths are able to reduce lot of xenobiotics including some anthelmintics. In this project we would like to study the role of reduction biotransformation enzymes in decrease of anthelmintic efficacy against Haemonchus contortus in sheep (Ovis aries). Závažným celosvětovým problémem se stává léková rezistence červů - původců helmintóz hospodářských i volně žijících zvířat. V navození lékové rezistence helmintů může hrát významnou roli indukce (tj. zvýšení aktivity a exprese) biotransformační enzymů parazita i hostitele. Zatím v této souvislosti byly zkoumány pouze indukce oxidačních a konjugačních enzymů. Pozornost nebyla dosud věnována redukčním biotransformačním enzymům, ačkoli u parazitujících helmintů redukce představuje základní metabolickou přeměnu xenobiotik včetně některých anthelmintik. V navrženém projektu bychom chtěli studovat úlohu redukčních biotransformačních enzymů ve snížení účinnosti anthelmintik proti vlasovce slézové (Haemonchus contortus) parazitující u ovce domácí (Ovis aries). Cílem projektu je stanovit aktivity redukčních biotransformačních enzymů ovce domácí i vlasovky slézové vůči anthelmintiku flubendazolu a specifickým modelovým substrátům, studovat změny v aktivitě těchto enzymů v závislosti na léčbě ovce domácí
