We propose to study the cell-cell and cell-extracellular matrix interactions involved in corneal graft tolerance or rejection by means of detection of cytokine receptors, adhesion molecules and other immunoregulatory molecules in corneal grafts. The regulatory role of stimulatory and inhibitory cytokines will be determined on the basis of changes in levels in the anterior chamber fluid (ELISA assays). All experiments will be performed on murine model of orthotopic corneal transplantation. The typing of phenotypically and functionally distinct cellular subpopulations will be studied by means of flow cytometry and immunohistology. We will concentrate on the differences in rejection mechanisms involved in risk recipients with prevascularized corneas and unmodified recipients. Results will be compared in rejected and accepted corneas , obtained from both untreated animals and from recipients treated with immunosuppressive drugs. Ke studiu buněčných a molekulárních mechanismů rejekce a tolerance využijeme expermentální model orthotopické transplantace rohovky na geneticky definovaných inbredních myších kmenech. Na základě exprese adhezivních molekul, receptorů pro cytokiny, hladiny cytokinů a dalších imunoregulačních molekul v rohovkách, předně komorové tekutině a v regionálních uzlinách příjemců (imunohistologie, průtokové cytometrie, ELISA testy) budeme studovat mezibuněčné interakce a interakce mezi buňkami a extracelulární matrix po transplantaci alogenní rohovky. Hodnotit budeme změny ve sledovaných parametrech u neléčených a u imunosuprimovaných příjemců, kteří rejekovali nebo tolerovali transplantát. Provedeme analýzu mechanismů rejekce u nemodifikovaných příjemců a u rizikových příjemců s prevaskularizovanou rohovkou.
