Description
  • In the period 1998 to 1999 we initiated research of the cytoskeleton of the human patogenic yeasts at Chiba University Japan and this research continued in the following 5 years supported by  Czech Grant Agency (grants No. 310/00/0391and 310/03/1195). Weidentified cytoskeleton in the life cycles of Cryptococcus neoformans, C. laurentii, Malassezia pachydermatis, Fellomyces fuzhouensis and Aureobasidium pullulans and we found out that some cytoskeletal inhibitors cease the growth of pathogens in vitro. The aim of the present project is (i) to extend cytoskeleton research to dimophic yeasts (Candida albicans, Sporothrix schenkii and Paracoccidioides brasiliensis) whose morphological forms display a different pathogenity and (ii) to find out, whether cytoskeleton inhibitors or their combinations will selectively block some of the morphological forms of dimorphic yeasts (filamentous, yeast, conidia).   • V letech 1998-9 jsme na Chiba University Japan zahájili výzkum cytoskeletu lidských patogenních kvasinek a v letech  2000-2005 tento výzkum pokračoval na domácím pracovišti za podpory GAČR (granty č. 310/00/0391 a 310/03/1195). Jednak  jsme identifikovali cytoskelet v životních cyklech  Cryptococcus neoformans, Cryptococcus laurentii, Malassezia pachydermatis, Fellomyces fuzhouensis  a Aureobasidium pullulans a jednak jsme zjistili, že in vitro některé inhibitory cytoskeletu zastavují růst patogenů. Cílem současného projektu je (i) rozšířit výzkum cytoskeletu na dimorfní kvasinky (Candida albicans, Sporothrix schenkii a Paracoccidioides brasiliensis) jejichž morfologické formy mají rozdílnou patogenitu a (ii) zjistit, zda inhibitory cytoskeletu a/nebo jejich kombinace budou působit selektivně na některý z morfologických typů dimorfních kvasinek (vláknitý,kvasinkový, konidie). iii) Poněvadž mikrotubuly i F-aktin se účastní konidiogenese, pučení i růstu hyf (Kopecká, Gabriel et al.,
