Description
| - The gram+ bacterial genus Streptomyces is characterized by complex morphological differentation resembling that of filamentous fungi and the ability to produce a wide variety of secondary metabolites. Involvement of protein phosphorylation by eukaryotic-like protein kinases in the regulation of secondary metabolism in Streptomyces has recently been reported. We have described the presence of gene family coding for protein Ser/Thr kinases in S. granaticolor and two of them were cloned and their sequenceshave already been determined. During this project, other genes belonging to the family will be sequenced and further characterized. Sites of transcription initiation and the level of expression during development will be analyzed. Properties of the puri fied enzymes will be studied both in vitro and in vivo. The chromosomal protein kinase genes will be disrupted and the effect of gene silencing will be studied. - Streptomycety jsou G+ bakterie, pro které je charakteristická morfologická diferenciace a produkce sekundárních metabolitů. Nedávno bylo zjištěno, že regulace sekundárního metabolismu probíhá mimo jiné na úrovni fosforylace proteinů, která je zajišťovánaprotein kinázami eukaryontního typu. U Streptomyces granaticolor jsme prokázali přítomnost genové rodiny Ser/Thr protein kináz, přičemž dva geny již byly klonovány a určena jejich sekvence. V navrhovaném projektu máme v úmyslu charakterizovat další homo logní geny a určit transkripční signály a hladinu exprese během diferenciace, určit vlastnosti purifikovaných enzymů in vitro a in vivo, inaktivovat chromozomální geny kódující protein kinázy a studovat vliv inaktivace na fyziologii a globální metabolismus. Protilátky proti purifikovaným enzymům nám umožní studovat jejich interakci se substráty. Označené purifikované proteiny budou použity jako sondy při detekci klonů v expresní knihovně kódujících substráty protein kináz. Interagující bílkoviny budeme
