Description
  • Disp3 is a sterol-sensing domain-containing protein that is related to the family of Dispatched proteins. Expression profiling of Disp3 revealed predominant expression in the brain and retina. DISP3 was found to co-localize with cholesterol in the ER and upon overexpression induced lipid droplet formation. We have identified two neural stem cell lines that highly express Disp3. Disp3 expression is positively regulated by T3 treatment and upon differentiation the level of Disp3 dramatically changes suggesting Disp3 may modulate self-renewal or differentiation. Brain tumors such as medulloblastoma are believed to arise from neural precursor cells. Analysis of a small number of primary tumors revealed very high expression of Disp3 suggesting this protein is important for their pathogenesis. We will conduct RNAi and overexpression studies to determine the role of Disp3 in neural stem cell maintenance. We will also create Disp3 transgenic mice to determine the neural specific effects of Disp3 overexpression on lipid homeostasis and the onset of neurodegenerative disease. (en)
