The aim of this project is to contribute to an elucidation of the viral infection mechanism during individual steps of viral infection and to the functional determination of individual proteins, encoded by viral genome, during infection process. The project is directed to two major research areas. The first one represents the ultrastructural analysis of polyoma virion movement in the host cell during individual steps of infection process and determination of cellular proteins which cooperate with viral components during this infection. The second area is devoted to investigation of cell morphology changes after inducible expression of individual viral genes and to study of viral components interaction with cytoskeletal and nucear matrix proteins.The results of the project can serve as a basis for developing of host protection, targeting of antiviral and antioncogenic agents and for developing of efficient delivery system for gene therapy. Předkládaný projekt si klade za cíl přispět k objasnění mechanizmu virové infekce polyomavirem během jejích jednotlivých fází a k určení funkčních vlastností proteinů kodovaných virovým genomem. Projekt je směrován do dvou hlavních oblastí. Prvá oblast zahrnuje ultrastrukturní analýzu pohybu částic polyomaviru v hostitelských buňkách během jednotlivých fází infekčního procesu a nalezení těch buněčných proteinů, které jsou v tomto procesu funkčně spojeny s virovými složkami. Druhá oblast je věnována sledování morfologických změn buněčných struktur po indukované expresi jednotlivých virových genů a studiu interakce virových složek s proteiny cytoskeletu a jaderné matrix. Získané výsledky mohou sloužit jako podklad k vypracování strategie obrany hostitelských buněk, pro směrování antivirových a antionkogenních látek a zároveň pro přípravu účinného systému pro účelu genové terapie.
Projekt byl řešen na velmi vysoké vědecké úrovni a přinesl řadu originálních výsledků v oblasti transportu polyomavirů uvnitř histotelské buňky. Bylo prokázáno, že polyomavirus i VP1 pseudokapsidy vstupují do buněk v monopinocytických váčcích, proteiny k (cs)
