Temperature sensitive (Ts) mutants of thermophilic proteins are powerful tool for studying protein function and stability. We propose a method to predict those residues in a given protein that, when appropriately mutated, are most likely to give rise toa Ts phenotype or to change temperature optimum. The aim of the study is to indicate a set of hydrophobic/aromatic amino acid substitutions that should be made to generate a Ts mutant of the protein. We are going to explore the relation between the hydrophobic-hydrophobic interactions in the protein core and the protein stability by an analysis the optimized hydrophobic-hydrophobic interactions. The fundamental importance of this project is that using a combination of theoretical calculations and experimental data determined at laboratory of applicant or from literature the relationship between these interactions and the protein stability is explored. Teplotně senzitivní mutanty termofilních proteinů jsou významným zdrojem informací pro studium stability a funkce proteinů. V předloženém projektu popisujeme metodu která by měla umožnit identifikaci těch aminokyselin jejichž mutací dojde ke změně fenotypu nebo ke změně teplotního optima. Cílem práce je identifikace řady hydrofobních/aromatických aminokyselin jejichž změnou jsme schopni požadované funkce dosáhnout. Tato identifikace bude probíhat studiem vztahů mezi hydrofobními aminokyselinami vproteinovém core a celkovou stabilitou proteinu a analýzou optimalizovaných hydrofobních interakcí. Základním prvkem této studie je korelace výsledků získaných rozvinutými výpočetními metodami kvantové chemie na modelových systémech a experimentálních údajů změřených na řešitelském pracovišti nebo z literatury popisujících proteinovou stabilitu termodynamicky. Z praktického hlediska je využití tohoto přístupu aplikovatelné k návrhu proteinů stabilních při vysokých teplotách.
