Podstatou tohoto projektu je výzkum vlivu konformace DNA na katalytickou účinnost DNA-dependentních DNA a RNA polymeráz s využitím templátů DNA obsahujících místně specifické adukty různých protinádorově účinných sloučenin platiny a ruthenia a jejich modelových analogů. Dlouhodobé cíle takto orientovaného výzkumu jsou porozumět mechanismům rezistence u farmak odvozených od komplexů kovů a jejich protinádorovým a mutagenním efektům. Kromě toho metodologie založené na využití DNA a RNA polymeráz na templátech DNA globálně modifikovaných protinádorově účinnými sloučeninami platiny a ruthenia jsou využívány při replikačním a transkripčním mapování míst v DNA, ve kterých dochází k zastavení replikace nebo transkripce DNA a tedy také míst, ke kterým se v DNA preferenčně váží protinádorově účinné komplexy kovů. Dalším dlouhodobým cílem našeho výzkumu je tedy také zdokonalit teoretické zázemí pro replikační a transkripční mapování míst v DNA poškozených biologicky významnými látkami.
This project proposes to examine the effect of DNA conformation on the catalytic efficiency and fidelity of DNA-dependent DNA and RNA polymerases on DNA templates containing site-specific adducts of various antitumor platinum and ruthenium compounds and their model analogs. The long-term goals are to understand the mechanism underlying resistance to metal-based drugs and their antitumor and mutagenic affects. In addition, the methodology based on the use of DNA and RNA polymerases and DNA templates globally modified by platinum or ruthenium antitumor compounds has been used in the %22replication%22 or %22transcription%22 mapping experiments to identify the arrest sites of DNA replication or transcription and consequently also preferential DNA binding sites of antitumor metal-based compounds. Thus, another long-term goal of our research is to improve theoretical background for the replication and transcription mapping of the lesions formed in DNA by the compounds of biological significance.
