Description	The objective of the project proposal is analysis of organometalic compounds – biological active ligands of retinoid X receptors. The specific aim of this study will be investigation of molecular mechanisms in relation to therapy of human breast cancer. In the course of whole duration of research, we will continually accomplish analyses of nuclear retinoid X receptor subtypes (RXRalpha, RXRbeta, RXRgamma) expression, retinoic acid receptor subtypes (RARalpha, RARbeta, RARgamma) expression and expression of selected corepressors (N-CoR, SMRT) and coactivator (SRC-1) in MCF-7 breast cancer cell line. Proteomic analysis aimed at the evaluation of process and efficiency of biological active ligands treatment will be proceed within the project. Monitoring of mentioned ligands effect will be evaluated on the basis of proteins profile changes in samples obtained during the project. (en) Cílem předkládaného projektu je analýza organokovových biologicky aktivních ligandů retinoid X nukleárních receptorů s cílem studia jejich molekulárních mechanismů ve vztahu k terapii lidských karcinomů prsu. V průběhu celého období řešení projektu se budou kontinuálně uskutečňovat analýzy exprese nukleárních receptorů podtypů retinoid X receptorů (RXRalfa, RXRbeta, RXRgama), exprese podtypů retionidních receptorů (RARalfa, RARbeta, RARgama) a některých jejich korepresorů (NCoR, SMRT) a koaktivátoru (SRC-1) v nádorové linii prsních buněk MCF-7 na základě vlivu perspektivních ligandů RXR nukleárních receptorů. Budou provedeny proteomické analýzy s cílem vyhodnotit průběh a efektivnost působení biologicky aktivních ligandů. Sledování účinku zmiňovaných ligandů bude vyhodnocováno na základě změn v proteinovém zastoupení ve vzorcích získaných během celého projektu.
