  • Selection agents will be derived from serious pathogens of peas (Fusarium solani f.sp. pisi, Mycosphaerella pinodes), faba bean (F. solani, f. sp. viciae fabae, Ascochyta fabae), and lupin (F. solani, f.sp. lupini, Colletotrichum gleosporioides), aimingto their suitability for utilisation in selections of embryogenic and organogenic cultures in vitro. The three types of selective agents (crude autoclaved filtrate, consequently filtered solution of metabolites, elicitors derived from fungal cell wall components) will be searched for their effects in the in vitro cultures (morphological and physiological changes, content of pathogenesis-related proteins). The reactions in vitro will be compared to the effects of inoculations in vivo. The lines selectedfrom in vitro cultures will be analysed for the content of PR proteins, polymorfism of isoenzymes and DNA and DNA content. The resistance of selected regenerants will be assessed in inoculation tests. (en)
  • U závažných patogenů hrachu (Fusarium solani f.sp. pisi, Mycosphaerella pinodes), bobu (F. solani, f. sp. viciae fabae, Ascochyta fabae) a lupiny (F. solani, f.sp. lupini, Colletotrichum gleosporioides) budou odvozena selekční agens vhodná pro selekce vembryogenních a organogenních kulturách in vitro. U tří typů selekčních agens - hrubý autoklávovaný filtrát, postupně filtrovaný roztok metabolitů a elicitory izolované z buněčných stěn hub - bude sledován účinek v kulturách in vitro (morfologické a fyziologické změny, obsah pathogenesis-related (PR) proteinů). Po srovnání reakce in vivo dosažené inokulacemi budou hledány korelační vztahy mezi reakcí in vitro a in vivo. Linie selektované in vitro budou analyzovány na obsah PR proteinů, polymorfismus isoenzymů a DNA a obsah DNA. Odolnost selektovaných regenerantů k patogenům bude hodnocena v inokulačních testech.
