  • To determine, by immunohistochemistry, glycosphingolipid localisation in the canalicular and sinusoidal membranes and localisation transfer to the sinusoids both in induced experimental cholestasis and in human biopsy and autopsy speciments. To monitor the effect of the induction agent in an effort to determine wheter transfer could mean an effort by the cell to reinforce sinusoidal pole towards increased blood bile acid levels. To test the reversibility of these changes.   • Imunohistochemicky prokazovat lokalizaci glykosfingolipidů v kanalikulární a sinusoidální membráně a přesun lokalizace do sinusoid jednak u indukované experimentální cholestázy i na lidských biopsiích a nekropsiích. Sledovat vliv indukčního agens a se snahou objasnit zda přesun by mohl znamenat snahu buňky o zpevnění sinusoidálního pólu vůči zvýšené koncentraci žlučových kyselin v krvi. Ověřit reversibilitu těchto změn. Studovat membránové mikrodomény obohacené na glykosfingolipidy a jejich roli v signalizaci a tvorbě membránových komplexů s využitím dvoukrokové solubilizace saponin /Triton X-100/. Studovat strukturu, dynamiku a funkci signalizačních komplexů po ovlivnění HEPG2 buněk interleukinem-6. (cs)
