  • Primárním cílem projektu je pochopení molekulární podstaty interakce bakteriální senzorové proteinkinázy s ligandem a vlivu této interakce na buněčné funkce, jež jsou na ní závislé. S tím souvisí i studium vlivu modifikace fosforylací jednotlivých substrátů signální dráhy na jejich enzymatickou funkci. (cs)
  • The direction of the project is aimed to solve the three-dimensional structure of the sensor domain of the unique bacterial receptor kinase and to elucidate the nature of its binding with the synthetic ligands. Furthermore, we anticipate to use site-directed mutagenesis and explore that substitution of the amino acids within the binding site for the ligands will result in sensor domain with diminished affinity or abrogated binding ability for the ligand molecules. Consequently, kinase-dependent signalling cascade will be blocked. The collective set of experiments should shed definitive light on bacterial intracellular signaling and on virulence. (en)
