| - Plant development is the result of an intricate spatial and temporal multiple hormonal control, many aspects of which are poorely understood. The project adresses many unanswered questions in the field of research of the two most important hormone groups, cytokinins (CKs) and auxins. It includes studies of biosynthesis of aromatic CKs; regulation of the level of free CKs by conjugation and/or hydrolysis of conjugates and by oxidation; in regulation of cell division, apical dominance and some in vitro systems (somatic embryogenesis, regeneration); mechanism of habituation and mechanism of signal transduction. In the frame of the project molecular techniques will be introduced into hormonal studies. The mentioned research requires reproducible anreliable determination of contents of CKs and auxins and their metabolites. The contemporary standard of hormone analyses requires mass spectrometric detection. That is why we are asking a GC-tandem MS equipment, which will be used to analyse hormones an (en)
- "Růst a vývoj rostlin je výsledkem složitých hormonálních interakcí, které jsou koordinovány v čase a prostoru. Jejich podstata není doposud uspokojivě objasněna. Projekt řeší mnoho otázek spojených s nejdůležitějšími skupinami fytohormonů - cytokininy (CK) a auxiny. Zahrnuje studium biosyntézy CK, regulaci hladiny volných CK prostřednictvím konjugátů, hydrolýzou konjugátů a biodegradací. Dále řeší vzájemné vztahy světla k transportu i metabolismu auxinů a CK, roli auxinů a CK v regulaci buněčného dělení, v aplikální dominanci a v ""in vitro"" systému (somatická embryogenese, regenerace), mechanismus habituace a mechanismus přenosu hormonálních signálů. V rámci projektu budou použity v hormonálních studiích molekulární techniky. Zamýšlený výzkum vyžaduje reprodukovatelné stanovení obsahu CK a auxinů a jejich metabolitů. Současné standartní hormonální analýzy vyžadují odpovídající GS-MS detekci, proto požadujeme GS-tandem zařízení, které bude analyzovat hormony a jejich metabolity pro všechny pra" (cs)
