Description
| - Enzym FerB byl naší skupinou zařazen do rodiny NADPH-dependentních FMN reduktas, redukující kromě Fe(III)komplexů i chromany a především chinony. FerB redukuje benzochinony dvouelektronově, naftochinony a chromát mechanismem jednoelektronovým za produkce reaktivních forem kyslíku. Rekombinantní enzym má stejné enzymové aktivity jako vlastní protein FerB a navíc snadno krystalizuje, což jsou dvě nezbytné podmínky pro další strukturně-funkční studie, které navrhujeme v předkládaném projektu s proteinem provést. Vyřešení 3D struktury celého proteinu umožní určit uspořádání aminokyselin v aktivním místě a následná evoluční studie by měla vést k pochopení důvodu dvojakosti samotného reakčního mechanismu a způsobu, kterým jsou radikály generovány. Aminokyseliny, které hrají při redukci substrátu největší roli, budou mutovány. Nejlépe pozměněný protein bude charakterizován s ohledem na jeho nové strukturní a funkční vlastnosti. Bude testováno možné využití biosenzoru s imobilizovaným rFerB. (cs)
- The enzyme FerB was included by our group into the family of NADPH-dependent FMN reductase reducing except Fe(III)complexes even chromate and especially quinones. FerB reduces benzoquinones by two-electron process, naphthoquinones and chromate by one-electron mechanism resulting in the production of oxygen reactive forms. Recombinant enzyme has same enzyme activities like own protein FerB and is easily crystallized, what are two necessary conditions for further structural and functional studies that we propose to make in the project. Solving of the 3D structure of the whole protein allows determining an arrangement of amino acids in the active site and subsequent evolution study should lead to understanding of double route reaction mechanism and the way how radicals are generated. Amino acids that are playing the most important role during the substrate reduction, will be mutated. The best bioengineered protein will be characterized in the view of new structural and functional features. Possible usage of the biosensor with immobilized rFerB will be tested. (en)
