Jak fungují fotoperiodické hodiny na molekulární úrovni a zda fotoperiodické a cirkadiánní hodiny sdílejí společné molekulární komponenty, není jasné. Jelikož současné modelové organismy jsou nevhodné pro studium fotoperiodických hodin, zvolili jsme ruměnici pospolnou, která vykazuje silnou diapauzní odpověď na fotoperiodu, a u které již existují nediapauzní kmeny. Naším cílem je zmapovat mutace u těchto kmenů, což by mělo přispět k pochopení podstaty fotoperiodických hodin. Naše současné výsledky naznačují účast některých circadiánních produktů v regulaci diapauzy. Proto provedeme lokalizaci a kvantifikaci těchto produktů u mutantních linií a divokého kmene a zjistíme vliv fotoperiodických a teplotních podmínek na jejich expresi. (cs)
Although seasonal clocks are widespread in animals, their molecular mechanism is not understood. The key reason for this is the absence of solid photoperiodic phenotype in typical genetic model organisms. Therefore we will apply genetic methods and molecular to Pyrrhocoris apterus, to map mutation in our two non-diapausing strains. Our preliminary data already suggest that circadian gene products are involved in diapause regulation. Therefore we will asses the temporal and tissue expression patternof these molecules in wild-type and mutant animals, in context of conditions that either promote or block the organism's photoperiodic response.
