"Previously we characterized an oligofamily chalconsynthase genes chs_H1 from hop, expressed in glandular tissue and involved in production of ""medicinal"" prenylflavonoids and bitter acids important in brewing idustry. We also initialized an analysis of regulatory genes and characterized first Myb regulatory factors from hop using sequence homology approach. In the project proposed we aim to study a complex set of transcription factors (TFs) from R2R3Myb, bHLH and bZIP families, selected according to chs_H1 promoter binding sites.TFs will be isolated from cDNA libraries using tissue-specific probes and by means of promoter binding and protein:protein interaction systems. TFs interactions in vitro and in planta effects on metabolome will be analyzed in heterologous transgenotes and after transient expression in agroinfiltrated leaves. Quantification of TFs expression and sequence diversity analyses will be performed to compare high, and low-alpha hop genotypes. It is expected that the project contribute to analysis of hop metabolome regulation and TF-biotechnology." (en)
Hlavními cíli projektu je izolovat, sekvenčně charakterizovat a funkčně analyzovat v heterologních systémech soubor transkripčních regulačních faktorů chmelu zejména z genových rodin Myb, bZIP a bHLH majících vztah k biosyntéze lupulinu. (cs)
